Polymorphism of POU1F1 gene in goat by polymerase chain reaction-restriction fragment length polymorphism

POU1F1 gene is a member of the POU-domain family. It is a positive regulator for growth hormone, prolactin and TSH-β. This work aimed to study polymorphism of POU1F1 gene in Sirohi goat breed. Blood samples were obtained from 50 goats for the extraction of DNA. PCR was used to amplify 563-bp fragment from exon 3 and flankingb region of POU1F1 gene using a published primer pair. POU1F1 gene was genotyped by restriction enzyme TaaI using PCR-RFLP method. PCR-RFLP results revealed two alleles (G and T) and three genotypes GG, TT and GT respectively. Polymorphism POU1F1-TaaI expresses significant gene and genotype frequencies. Maximum gene frequency was observed for G (0.55). Maximum genotype frequency was observed for GG 0.40 whereas TT and GT genotype showed equal genotype frequency of 0.30.
